Tieto zariadenia vyu\u017e\u00edvaj\u00fa anizotropiu optick\u00fdch, elektrick\u00fdch a magnetick\u00fdch vlastnost\u00ed kvapaln\u00fdch kry\u0161t\u00e1lov. Projekt sa zameriava na sk\u00famanie \u0161trukt\u00farnych zmien vyvolan\u00fdch vonkaj\u0161\u00edmi po\u013eami v kvapaln\u00fdch kry\u0161t\u00e1loch dopovan\u00fdch magnetick\u00fdmi, nemagnetick\u00fdmi a fototerm\u00e1lnymi nano\u010dasticami ako aj ich kombin\u00e1ciou. Tieto \u0161trukt\u00farne zmeny zah\u0155\u0148aj\u00fa f\u00e1zov\u00e9 a orienta\u010dn\u00e9 prechody tak\u00fdchto kompozitn\u00fdch syst\u00e9mov. Tento projekt je zameran\u00fd na vyu\u017eitie prote\u00ednov neskorej embryogen\u00e9zy (LEAPs) z\u00edskan\u00fdch z rastl\u00edn schopn\u00fdch regener\u00e1cie, ako nov\u00fdch anti-agrega\u010dn\u00fdch l\u00e1tok pre k\u013e\u00fa\u010dov\u00e9 prote\u00edny spojen\u00e9 s patogen\u00e9zou AD a PD, amyloid \u03b2 peptid a \u03b1-synukle\u00edn. Na\u0161e predbe\u017en\u00e9 v\u00fdsledky uk\u00e1zali, \u017ee LEAPs inhibuj\u00fa amyloidn\u00fa agreg\u00e1ciu in vitro, \u010do bolo potvrden\u00e9 pokro\u010dil\u00fdmi spektroskopick\u00fdmi a zobrazovac\u00edmi technikami. Cie\u013eom predkladan\u00e9ho projektu bude rekombinantne pripravi\u0165 LEAPs, objasni\u0165 ich anti-agrega\u010dn\u00fd potenci\u00e1l vo\u010di A\u03b240 peptidu a \u03b1-Syn a vyvin\u00fa\u0165 LEA-mimetick\u00e9 peptidy s vy\u0161\u0161\u00edm terapeutick\u00fdm potenci\u00e1lom. Prostredn\u00edctvom in vitro met\u00f3d, neur\u00e1lnych bunkov\u00fdch modelov a in vivo modelov AD\/PD-C. elegans, budeme hodnoti\u0165 ich anti-agrega\u010dn\u00fa \u00fa\u010dinnos\u0165 a cytotoxicitu. Amyloidn\u00e9 prote\u00edny s\u00fa kateg\u00f3riou programovate\u013en\u00fdch samo-sa usporiadavaj\u00facich makromolek\u00fal a v\u010faka t\u00fdmto vlastnostiam je mo\u017en\u00e9 racion\u00e1lne manipulova\u0165 a programova\u0165 vznikaj\u00face \u0161trukt\u00fary. Samousporiadanie m\u00f4\u017ee navy\u0161e prebieha\u0165 aj pri kombin\u00e1cii viacer\u00fdch stavebn\u00fdch blokov (typov makromolek\u00fal), \u010do vedie k vzniku \u0161truktur\u00e1lne zlo\u017eitej\u0161\u00edch zost\u00e1v, ktor\u00e9 m\u00f4\u017eu ma\u0165 odli\u0161n\u00e9 fyzik\u00e1lno-chemick\u00e9 vlastnosti od vlastnost\u00ed p\u00f4vodn\u00fdch mono\u0161trukt\u00far. Preto v r\u00e1mci strat\u00e9gie riaden\u00e9ho samousporiadania a zosie\u0165ovania je mo\u017en\u00e9 mal\u00fdmi \u00fapravami v reak\u010dn\u00fdch podmienkach kontrolova\u0165 v\u00fdsledn\u00e9 fyzik\u00e1lno-chemick\u00e9 vlastnosti hydrog\u00e9lov na b\u00e1ze amyloidn\u00fdch vl\u00e1kien. SUSY \u010dast\u00edc a s nimi sp\u00e4tej te\u00f3rie supersymetrie a napokon je to anal\u00fdza zameran\u00e1 na sk\u00famanie smerov fyziky mimo r\u00e1mca \u0160M, ako je napr. na mo\u017enos\u0165 existencie extra dimenzi\u00ed, mikro \u010diernych dier a podobne. Napriek odli\u0161n\u00fdm sympt\u00f3mom, NDD zdie\u013eaj\u00fa nieko\u013eko spolo\u010dn\u00fdch patog\u00e9nnych mechanizmov.V s\u00fa\u010dasnosti neexistuje liek pre tieto ochorenia a komplexnos\u0165 ich patol\u00f3gie si vy\u017eaduje inovat\u00edvne terapeutick\u00e9rie\u0161enia. V tomto projekte bude pou\u017eit\u00e1 nov\u00e1 strat\u00e9gia multi-cie\u013eov\u00fdch ligandov (MTDL), ktor\u00e1 sa zameriava nav\u00fdvoj nov\u00fdch zl\u00fa\u010den\u00edn moduluj\u00facich viacero procesov spojen\u00fdch s NDD s\u00fa\u010dasne. Zameriame sa na \u010d\u00ednske byliny \u2013Angelica sinensis, Ligusticum chuanxiong, a Cinnamomum cassia. Cie\u013eom projektu je presk\u00fama\u0165 anti -agrega\u010dn\u00fdpotenci\u00e1l t\u00fdchto rastlinn\u00fdch extraktov a ich deriv\u00e1tov na A\u03b2 peptid aj \u03b1 -synukle\u00edn, pri\u010dom by mali vykazova\u0165 ajneuroprotekt\u00edvne a protiz\u00e1palov\u00e9 vlastnosti. Projekt kombinuje odborn\u00e9 znalosti a sk\u00fasenosti oboch v\u00fdskumn\u00fdcht\u00edmov v oblasti. Nespr\u00e1vneho zba\u013eovanie prote\u00ednov a patol\u00f3gie NDD, a umo\u017en\u00ed n\u00e1m z\u00edska\u0165 komplexn\u00e9 \u00fadajepomocou komplement\u00e1rnych pr\u00edstupov, \u010do bude vies\u0165 k n\u00e1vrhu mo\u017en\u00fdch terapeutick\u00fdch alternat\u00edv proti t\u00fdmtoochoreniam.
